Кріопошкодження цілісності ДНК сперматозоїдів свійських тварин: методи визначення і шляхи підвищення цілісності ДНК

Анотація

У огляді представлено дані щодо впливу кріоконсервування на стан ДНК сперматозоїдів свійських тварин, описано основні причини фрагментації ДНК, методи визначення та способи збереження її цілісності. Показано, що наявні методи визначення фрагментації ДНК є досить ефективними, але вибір оптимального методу обумовлений видовою приналежністю тварини. Основним чинником кріопошкоджень ДНК визнано оксидативний стрес, спричинений підвищеним вмістом активних форм кисню. Для запобігання даного ефекту розроблено багато захисних середовищ та реабілітаційних розчинів з додаванням антиоксидантів (неферментні, ферментні сполуки та наночастинки). Таким чином, визначення рівня фрагментації ДНК сперматозоїдів та збереження її цілісності є необхідною складовою для підвищення ефективності кріоконсервування гамет свійських тварин, що є складовою репродуктивних технологій.
